Two things I would love to see, more than anything:
1. An API to programmatically talk to Synfire
2. Open file formats
This would allow for third parties to come in and create bridges between DAWs and Synfire, create their own custom workflow solutions, etc. And then Synfire can focus on what it does best ("crunching the numbers"), without limiting the workflow possibilities.
Of course I do realize designing an API is a very non-trivial thing. But then again, Synfire is also a very expensive product! ;-)

I'll definitely throw in my request for a file format to exchange data to and from Synfire. Would be a real lifesaver.. :)
MIDI doesn't work so well. I'd love to be able to hint on chord changes and such for a part (different keyfinding algorithms)...or express the intervals explicitly for some input data without having to analyze and edit manually.
On the same hand, a batch import (even if it is mostly automatic settings) would also be useful. Especially for things like bulk MIDI drum groove imports. Those can be too time-consuming to import as it is.
