Harmony Navigator can not convert audio signals to MIDI notes (MIDI is what you need to feed HN with).

There are several software utilities that claim they can do this, but I doubt this works for complex music mixes. In most cases this requires a simple mono instrument (flute, for instance) to get reliable results.

The easiest way to get a melody into HN (or any other software) is to play it on a keyboard or MIDI guitar. Or you could transcribe it note by note with a sequencer and export that as a midi file. Vocals are usually simple, so that might perhaps be a viable solution for you.

Once you imported the melody into HN, it can help you find suitable chords using the harmonizer.
